He has agreed to a four-year contract with the north London club and will join once a work visa has been granted.
After goalkeeper Fraser Forster and winger Ivan Perisic, Bissouma, 25, becomes Antonio Conte’s third acquisition of the summer.
The Mali international’s Brighton contract was scheduled to expire next summer, and he was garnering interest from Aston Villa, who tried to sign him during the winter transfer window.
After finishing fourth last season, Spurs made a move and were able to give him the opportunity to play Champions League football.
Brighton manager Graham Potter said: “The transfer is a very good one for the club and player.
“Yves has shown his undoubted ability and been an important player during his time with us.
“Now he has a new challenge with Tottenham, including the opportunity to play Champions League football, and everyone at the club wishes him well.”
Bissouma scored one goal in 26 league games for Graham Potter’s club in 2021-22. He was a force in the center of their midfield as they reached ninth in the standings, the club’s greatest ever finish.
